The Overall Process of Gene Cloning
Generally, gene cloning involve 4 major steps, which are :
Each of these major steps will be further divided into many smaller steps and they will be discussed in the page followed. The picture on the left shows the basic concepts of how a recombinant DNA is formed and obtained. Basically, a foreign DNA with gene of interest is cut from the genome of an organism. The vector with the similar restriction site will be cut using the same restriction enzyme as the one used to cut the DNA fragment. Then, the DNA fragment will be inserted to the vector using ligase. After that, the recombinant DNA will be transform into bacteria (host). Finally, the bacteria will udergo screening to select the bacteria that are sucessfully recombinant. |
